I gather it's the person's first marriage, but not sure what the "6E" (?) bit means and why they mention "first" twice. Does it perhaps mention "after 6 weeks" of banns or whatever?

It means they are both marrying for the first time. The wording is rather abbreviated here.

A typical form of wording is: τέλεσε πρώτο γάμο σε πρώτο σύμφωνα με όλους τους τύπους ...
Of course, "πρώτο γάμο σε πρώτο", in itself, doesn't seem to make much sense, but other marriage certificates use a longer form of wording which is easier to understand.

Here's a more comprehensible version:
τέλεσε Α' πρώτο γάμο πολιτικό με την σε Β' δεύτερο γάμο συνερχόμενης (+ bride's name)
... and of course you'll have to expand the wording in English, with the words "this being her first marriage" or something on those lines.
It's absolutely standard for these certificates to specify whether it's his first or second etc. marriage, and her first or second...

It should be συνερχόμενη above!
In your case, "μετά σε (Α) πρώτο", it looks as if they are using the vestiges of the older form of wording, "μετά .... συνερχομένης", without having enough room to write all the words in.
