The safety of SOF was also demonstrated in Phase 3 clinical trials (NEUTRINO, FISSION, POSITRON, FUSION and VALENCE). No adverse reactions specific to SOF were identified from these clinical studies, where SOF was well tolerated. The safety profile of SOF in HCV/HIV coinfected patients in PHOTON-1 and in patients awaiting liver transplantation in Study 2025 was similar to that observed in subjects treated with SOF+RBV in Phase 3 clinical trials. Sovaldi has demonstrated high efficacy, acceptable tolerability and safety profiles to support use in patients with HCV infection > 12 years of age. Clinical and postmarketing experience of up to 210,487 patient-years exposure has demonstrated a favorable safety profile of SOF. During postmarketing experience, important identified risks of bradyarrhythmia when SOF is used with another DAA and amiodarone and HBV reactivation in HBV/HCV coinfected patients have been observed. The current SOF EU SmPC contains appropriate warnings and precautions regarding these risks. The important identified risks, as well as important potential risks for SOF, as described in the SOF EU-RMP, are summarized in Sections 16.4.1 and 16.4.2 respectively.

During the period of this PSUR/PBRER and in the context of the 5-year marketing authorization renewal for the product in the EU, the SOF EU-RMP was updated to meet the requirements of the revision to GVP Module V (EMA/838713/2011; Rev 2) and the RMP template (EMA/PRAC/613102/2015 Rev.2). As a result of review of safety concerns, 2 Important Potential Risks and 2 Missing Information were removed as part of the renewal application –
EU-RMP Version 7.0; dated 19 July 2018:
• Cardiac arrhythmia (bradycardia) in particular when concomitantly used with DCV and other bradycardic medicines
• DDI with potent intestinal Pgp inducers
• Safety in children (< 12 years of age)

Безпека SOF також була продемонстрована в клінічних випробуваннях 3 фази (NEUTRINO, FISSION, POSITRON, FUSION і VALENCE). Не було виявлено побічних реакцій, специфічних для SOF, у цих клінічних дослідженнях, в яких SOF добре переносився. Важливі ідентифіковані ризики, а також важливі потенційні ризики для SOF, як описано в Європейському плані управління ризиками для софосбувіру (SOF EU-RMP), узагальнені в Розділах 16.4.1 та 16.4.2, відповідно.

Протягом звітного періоду цього PSUR/PBRER та в контексті поновлення 5-річного реєстраційного посвідчення на лікарський засіб у країнах ЄС, Європейський план управління ризиками для софосбувіру (SOF EU-RMP) було оновлено відповідно до вимог оновленого Модуля V Належної практики фармаконагляду (GVP), (EMA/838713/2011; версія 2) та типового Плану управління ризиками (RMP) (EMA/PRAC/613102/2015, версія 2). У результаті огляду проблем безпеки в плані EU-RMP, версія 7.0 від 19 липня 2018 року, який є частиною заяви про продовження терміну дії реєстраційного посвідчення, було усунуто 2 важливих потенційних ризика та 2 випадки відсутньої інформації:

• Серцева аритмія (брадикардія), зокрема, при одночасному застосуванні з DCV (даклатасвіром) та іншими брадисистолічними препаратами.
