原文文本 - English英语 The main reason why the major part of the experiments was carried out in anesthetized rats was the practical usefulness of this protocol, both in terms of speed and the ethical issues, when testing the effects of novel (antidepressant) drugs targeting all three monoamines. In this respect, the effects of the combined citalopram and methylphenidate treatment on extracellular monoamine levels may serve as “positive controls”.
Acute administration of SSRI citalopram caused marked increases in 5-HT concentrations in microdialysates from all brain structures examined, in agreement with earlier studies (Millan et al., 1999, 2001) showing the ceiling effect of citalopram administered at this dose on extracellular 5-HT levels in the rat brain. Accordingly, citalopram did not change the levels of NA and DA in the PFC and Nacc, while in the Hipp the overall deltaAUC value for DA was slightly higher in citalopram-treated anesthetized but not awake rats. Interestingly, a recent study demonstrated that citalopram, did not change the basal DA and NA overflow in the PFC but when given at a higher dose (10 mg/kg s.c.) it increased DA and NA concentrations in the parietal and occipital cortices (Valentini et al., 2005).
